Objective:To determine the pharmacokinetics and pharmacodynamics of sildenafil citrate in normal male subjects.
Design:A single-blind, randomized, single-dose crossover study design.
Patients and methods:Subjects received sildenafil citrate (25, 50, or 100 mg/day) or placebo as the active substance for the first six months of therapy.
Results:After six months of treatment with sildenafil citrate (25, 50, or 100 mg/day), mean total SFI1 (±SD) and mean total SFI2 (±SD) were 1.3 and 1.4% for sildenafil and placebo, respectively.
Conclusions:A single-dose, single-administration study design with a single-subject, single-dose, multiple-dose study design will be required to determine the pharmacokinetic and pharmacodynamics of sildenafil citrate in normal male subjects.
